ABSTRACT

Fungi are important in several aspects of human life. In particular, to agriculture, pathogenic fungi are of great importance, as they are responsible for production losses of the most diverse types. Because of this, knowledge about pathogenic fungus is of extreme importance for farmers and professionals working in agricultural areas. Among farmers who use specific agroecological practices, this knowledge is even more valuable, since by not adopting conventional methods of production, they resort to non-invasive alternatives that are less or not harmful at all to the environment in consideration of production management methods. This study aimed to assess farmer perception in the Cerrado biome in the city of Goiás (GO), Brazil, in order to understand their ethnomycological perceptions to verify historical management practices, their knowledge about phytopathogenic fungi, and how these producers perceive fungi. We used the theoretical reference method "From peasant to peasant" formulated by ANPA - National Association of Small Farmers. Some aspects of farmers' ethnomycological knowledge are discussed. These ease identification the representatives of the Fungi Kingdom is associated with organisms that present easily recognizable characteristics, such as wood-ears or disease-causing fungi. In general, farmers are able to identify representatives of the Fungi Kingdom that are found in their daily lives. The perception of farmers about fungi, a group still much unknown by society, is very relevant for future actions of ethnomycology.

ABSTRACT

This study aimed to identify Coagulase-Negative Staphylococci (CoNS) species isolated from bovine mastitis, through phenotypic and PCR-RFLP (Restriction Fragment Length Polymorphism-Polimerase Chain Reaction) methods and to compare both techniques to matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) technique. Among them, the PCR-RFLP method, using a partially conserved sequence of the groEL gene, is a promising alternative, because of its reproducibility and reliability. On the other hand, the proteomic technique MALDI-TOF MS provides an accurate and much faster diagnosis and has been increasingly employed in microbiological identification. The pheno-genotypic profiles of beta-lactam resistance were also investigated, this characterization is important, considering that the use of antimicrobials is a key element for mastitis control in dairy farms. The concordance of the phenotypic, PCR-RFLP and MALDI-TOF MS assays to identify CoNS species was 77,5% (31/40). S. chromogenes was the species most frequently isolated. Antibiotic resistance rate was relatively low, registering values of 25.5% to penicillin, 9.6% to oxacillin and 6.2% to cefoxitin. Resistance to imipenem, cephalotin and amoxicillin+clavulanate was not observed. The mecA gene and its variant were detected in 7.6% and 4,1% of the isolates respectively. The blaZ gene was found in 43.2% of the strains resistant to penicillin.(AU)

ABSTRACT

This study aims to detect the main virulence and antimicrobial resistance genes in Stapylococcus aureus from bovine mastitic milk as well as classifying them according to agr typing. A total of 55 strains from six dairy unities in the state of Rio de Janeiro were selected, of these 27.3% presented fbnA and 78,2% for fbnB genes, respectively. None of the strains tested were positive for cap5 gene, 3.6% were positive for cap8 gene. Additionally, 94.5% of strains had hlA gene and 89.1% had hlB gene while 67.3% of the strains had icaA gene and 87.3% had icaD gene. From these results it was possible to establish 12 different virulence profiles. Prevalence of agrII type was detected in 81.8% of the isolates. Concerning antimicrobial resistance evaluation, the studied strains were susceptible to all antibiotics tested except penicillin, 83.6% being resistant strains. None of the strains had mecA gene, however, 40% of the strains had blaZ gene. Associating virulence and resistance data made it possible to obtain 23 different profiles. This great diversity of strains shows wide array of bacterial strategies and the challenge of mastitis prevention in cattle. Despite antimicrobial susceptibility, these strains presented certain genes that allow its persistence in the herd.(AU)

ABSTRACT

The goals of this study were to identify microorganisms in dogs' ear canals through cytological and microbiological examination, and evaluate their association with otoacariasis and its clinical signs. Otodectes cynotis infestation diagnosis was achieved by bilateral otoscopy, and by parasite viewing on cerumem under stereoscopic microscope, representing the parasitological exam. Ear discharge was also collected from each canal to fix cytology slides and for microbiological isolation, but this last sample was collected with sterile swab. Among the 250 studied animals, 15 (6%) had ear mites. There was an association between the parasite and external clinical otitis specially as established by auricular erythema (P<0,001) and excessive production of ear discharge (P=0,0016), as well as ear pruritus observed by the owners (P<0,001). It has been suggested that these associations can be an effect of the infestations. There was also an association (P<0,01) between otoacariasis and microorganisms detected by cytological exam in both ear canals of each dog. However, there was no association (P=0,04 e P=0,07) between the presence of ear mite and microorganisms detected by microbiological exam of right and left ears.

ABSTRACT

The effects of the yeast cell wall (Saccharomyces cerevisiae) (CWSc) in the diet of broiler chickens were evaluated through the following treatments: reference diet with avilamicin (AV); reference diet (RD); RD with 0.1% CWSc (CWSc0,1); RD with 0.2% CWSc (CWSc0,2); RD with 0.3% CWSc (CWSc0,3). The study design was randomized blocks, with five treatments and six replicates of 10 birds, totaling 300 male broilers. In the period from 9 to 21 days treatment CWSc0.3 had a negative influence on weight gain (631g) and from 34 to 39 days CWSc0.1 provided better weight gain (461g) compared to the treatment with the antimicrobial. The highest live weight value at slaughter, 2,571 grams, was obtained in treatment CWSc0.1. The absolute weights of drumstick and thigh+drumstick were higher in treatments CWSc0.1 (290g and 560g) and CWSc0.2 (292g and 561g) and were lower for CWSc0.3 (263g and 515g). There was no influence of the treatments on both the metabolizability and the total count of coliforms in the ileum. Diets until 0.2% of PCSc inclusion resulted in correct productive rates and they can be used as a ration growth promoter additive in antimicrobial free diets without damage on performance, carcass characteristics, organ weight, metabolizability of the diets and total coliform count in the ileum.

ABSTRACT

Staphylococcus aureus is the major pathogen causing intramammary infections in dairy cattle worldwide. Among the factors that contribute to its spread and infectious potential is the ability to overcome the mechanisms of antimicrobials activity. The present work investigated the antimicrobial resistance pattern and sensibility to bacteriocins produced by strains of Lactobacillus spp of 30 isolates of S. aureus from mastitis. From this, 29 are beta-lactamase producers. Eight isolates (26.6 percent) showed resistance to at least four antibiotics being considered multiresistent. All of them were mecA-positive. Otherwise, all isolates tested showed sensibility to at least one of the four bacteriocin producer strains. Due to the significant depletion of the efficacy of antimicrobials, pathogen growth inhibition by bacteriocins seems an alternative of biological control in infectious processes.

ABSTRACT

Antimicrobials susceptibility pattern of bacterial isolated from different sites of infection, in vitro azithromycin activity pattern, and its minimum inhibitory concentration (MIC) values were evaluated. Different phenotypic tests detected azithromycin resistance in 45 percent of Staphylococcus spp. and 65 percent of resistant Gram-negative rods. MIC50 was 4.0μg/mL for Staphylococcus aureus, 1.0μg/mL for S. intermedius, >512.0μg/mL for coagulase negative Staphylococcus, and 256.0μg/mL for Gram-negative rods. In addition, it was investigated the possible cross-resistance between oxacillin and azithromycin, by detection of mecA gen in Staphylococcus spp. Nine (15 percent) mecA-positive Staphylococcus spp. were also phenotypically resistant to azithromycin.

ABSTRACT

A five-year retrospective study of dogs with patellar luxation was performed. A total of 75 dogs (n=122limbs) were studied: 82.7% (n=62) had medial luxation, 14.6% (n=11) had lateral luxation, and 2.7%(n=2) had both luxations in same stifle joint. Considering the dogs with medial patellar luxation, 6.7%(n=5) were mixed-breed and 76% (n=57) were purebred, with high prevalence in Poodles (n=27; 35.6%),Pinschers (n=13; 17.3%), and Yorkshires (n=4; 5.3%). Of the dogs with lateral patellar luxation, one(1.3%) was mixed-breed and 13.3% were purebred, with high prevalence in Poodles (n=6; 8%) andPinschers (n=2; 2.7%). Regarding the 62 dogs with medial patellar luxation, 20 were unilateral and 42bilateral. Of the 11 dogs with lateral patellar luxation, five were unilateral and six bilateral. Statistically, females were more affected than males by both medial and lateral patellar luxations. The grade II (25;33.3%) of patellar luxation was the most often in medial luxation followed by grades III (20; 26.7%), IV(11; 14.7%), and I (16; 7.9%). The grade II (5; 6.7%) was the most often in lateral luxation followed bygrade I (4; 5.3%), and simultaneously grades III (1.3%) and IV (1.3%). The surgical treatment wasperformed in 64 dogs and the others were submitted a conservative management. It was possible toconclude that among the dogs with patellar luxation evaluated in this study, the most affected were toyand miniature breeds and female dogs with medial patellar luxation.

ABSTRACT

ABSTRACT

The purpose of the present study was to investigate the expression (mRNA) of CD40 ligand (CD40L), interferon-gamma (IFN-gamma) and Fas ligand (FasL) genes in human cardiac allografts in relation to the occurrence of acute cardiac allograft rejection as well as its possible value in predicting acute rejection. The mRNA levels were determined by a semiquantitative reverse transcriptase-polymerase chain reaction method in 39 samples of endomyocardial biopsies obtained from 10 adult cardiac transplant recipients within the first six months after transplantation. Biopsies with ongoing acute rejection showed significantly higher CD40L, IFN-gamma and FasL mRNA expression than biopsies without rejection. The median values of mRNA expression in biopsies with and without rejection were 0.116 and zero for CD40L (P<0.003), 0.080 and zero for IFN-gamma (P<0.0009), and 0.156 and zero for FasL (P<0.002), respectively. In addition, the levels of IFN-gamma mRNA were significantly increased 7 to 15 days before the appearance of histological evidence of rejection (median of 0.086 in pre-rejection biopsies), i.e., they presented a predictive value. This study provides further evidence of heightened expression of immune activation genes during rejection and shows that some of these markers may present predictive value for the occurrence of acute rejection.

ABSTRACT

Rats implanted bilaterally with cannulae in the CA1 region of the dorsal hippocampus or the entorhinal cortex were submitted to either a one-trial inhibitory avoidance task, or to 5 min of habituation to an open field. Immediately after training, they received intrahippocampal or intraentorhinal 0.5-æl infusions of saline, of a vehicle (2 percent dimethylsulfoxide in saline), of the glutamatergic N-methyl-D-aspartate (NMDA) receptor antagonist 2-amino-5-phosphono pentanoic acid (AP5), of the protein kinase A inhibitor Rp-cAMPs (0.5 æg/side), of the calcium-calmodulin protein kinase II inhibitor KN-62, of the dopaminergic D1 antagonist SCH23390, or of the mitogen-activated protein kinase kinase inhibitor PD098059. Animals were tested in each task 24 h after training. Intrahippocampal KN-62 was amnestic for habituation; none of the other treatments had any effect on the retention of this task. In contrast, all of them strongly affected memory of the avoidance task. Intrahippocampal Rp-cAMPs, KN-62 and AP5, and intraentorhinal Rp-cAMPs, KN-62, PD098059 and SCH23390 caused retrograde amnesia. In view of the known actions of the treatments used, the present findings point to important biochemical differences in memory consolidation processes of the two tasks

ABSTRACT

Chick cardiomyocytes, when submitted to hyposmotic swelling, exhibit a partial regulatory volume decrease (RVD). A Ca2+ influx by stretch-activated channels signals a taurine efflux and the RVD at 37§C. We evaluated the cell's performance at room temperature. Cardiomyocytes isolated and cultured from 11-day-old chick embryos were submitted to a hyposmotic solution (180 mOsm/kg H2O) at 37§C and at room temperature (26§C). Under these conditions we measured the changes in cell volume as well as the intracellular free Ca2+ (using fura-2). During hyposmotic swelling, cells at 37§C displayed a peak relative volume of 1.61 + or - 0.03 and recovery to 1.22 + or - 0.04 (N = 14), while cells at 26§C presented a peak swell relative volume of 1.74 + or - 0.06 and did not recover (1.59 + or - 0.09, N = 9). Transient increases in intracellular Ca2+, which are characteristic of the normal RVD, were observed at both temperatures (29.1 + or - 4.5 percent (N = 8) and 115.2 + or - 42.8 percent (N = 5) increase at 37§ and 26§C (P<0.05), respectively). A delay in the Ca2+ transient increase was also observed when the cells were at 26§C (109 + or - 34 s compared to 38 + or - 9 s at 37§C, P<0.05). At room temperature the RVD does not occur because the calcium transient increase, which is an early event in the signaling of the RVD, is delayed. Also, free calcium is not cleared as in the 37§C RVD. In the normal RVD the free calcium returns to baseline levels. The very high and persistent free calcium levels seen at room temperature can lead to unregulated enzyme activities and may promote irreversible injury and cell death

ABSTRACT

Rats infected with the helminth Capillaria hepatica regularly develop septal hepatic fibrosis that may progress to cirrhosis in a relatively short time. Because of such characteristics, this experimental model was selected for testing drugs exhibiting antifibrosis potential, such as pentoxifylline, gadolinium chloride and vitamin A. Hepatic fibrosis was qualitatively and quantitatively evaluated in liver samples obtained by partial hepatectomy and at autopsy. The material was submitted to histological, biochemical and morphometric methods. A statistically significant reduction of fibrosis was obtained with pentoxifylline when administered intraperitoneally rather than intravenously. Gadolinium chloride showed moderate activity when administered prophylactically (before fibrosis had started), but showed a poor effect when fibrosis was well advanced. No modification of fibrosis was seen after vitamin A administration. Hydroxyproline content was correlated with morphometric measurements. The model appears to be adequate, since few animals die of the infection, fibrosis develops regularly in all animals, and the effects of different antifibrotic drugs and administration protocols can be easily detected
